| Assay Type | Activity: Active Biological Activity: Binding activity of Human Integrin Alpha-M (ITGAM) Protein (Active) measured using a functional ELISA. Recombinant Human Complement C3 (C3) protein was diluted serially in PBS with 0.01% BSA, pH 7.4. 100 μl replicates were then transferred to ITGAM-coated micro-wells and incubated for 1h at 37°C. Wells were washed with PBST and incubated for 1h with a polyclonal C3 antibody, then aspirated and washed 3 times. After incubation with a HRP-conjugated secondary antibody (1h at 37°C), wells were aspirated and washed 5 times. Substrate solution was added and wells were incubated for 15-25 minutes at 37°C. 50 μl stop solution was then added to the wells and read at 450/630 nm immediately. When recombinant Human ITGAM was immobilized at 2 μg/ml (100 ul/well), the concentration of recombinant Human C3 that produces 50% optimal binding was found to be approximately 0.130 μg/ml. Many proteins are synthesised as pro-proteins, and then cleaved to give the active form. 5. Polymerisation of the target protein. Dimerisation, multimerisation etc. will increase the band size observed. |
