Activity: Active
Biological Activity: Binding activity of Human Integrin Alpha-M (ITGAM) Protein (Active) measured using a functional ELISA. Recombinant Human Complement C3 (C3) protein was diluted serially in PBS with 0.01% BSA, pH 7.4. 100 μl replicates were then transferred to ITGAM-coated micro-wells and incubated for 1h at 37°C. Wells were washed with PBST and incubated for 1h with a polyclonal C3 antibody, then aspirated and washed 3 times. After incubation with a HRP-conjugated secondary antibody (1h at 37°C), wells were aspirated and washed 5 times. Substrate solution was added and wells were incubated for 15-25 minutes at 37°C. 50 μl stop solution was then added to the wells and read at 450/630 nm immediately. When recombinant Human ITGAM was immobilized at 2 μg/ml (100 ul/well), the concentration of recombinant Human C3 that produces 50% optimal binding was found to be approximately 0.130 μg/ml.

Calculated MW: 23.6 kDa  Observed MW (SDS-PAGE): 27 kDa, 30 kDa  Possible reasons why the actual band size differs from the predicted band size: 1. Splice variants. Alternative splicing may create different sized proteins from the same gene. 2. Relative charge. The composition of amino acids may affect the charge of the protein. 3. Post-translational modification. Phosphorylation, glycoslyation, methylation etc. may affect the band size. 4. Post-translational cleavage. Many proteins are synthesised as pro-proteins, and then cleaved to give the active form. 5. Polymerisation of the target protein. Dimerisation, multimerisation etc. will increase the band size observed.

To keep the original salt concentration, we recommend reconstituting to the original concentration prior to lyophilization (see Concentration) in ddH2O. If a lower concentration is required, dilute in 10 mM PBS, pH 7.4. If a higher concentration is required, the product can be reconstituted directly in 10 mM PBS, pH 7.4, though please note that this will change the overall salt concentration. The stock concentration should be between 0.1-1.0 mg/ml. Do not vortex.
