Descripción
ACKR2 is a non-classical chemokine receptor that functions as a decoy and scavenger receptor, specifically binding pro-inflammatory CC chemokines to modulate immune responses and resolve inflammation. Unlike signaling chemokine receptors, ACKR2 does not trigger intracellular signaling but instead internalizes and degrades its ligands, reducing chemokine availability and dampening inflammation. It is primarily expressed on lymphatic endothelial cells and plays a critical role in maintaining immune homeostasis by clearing chemokines from tissues and promoting the resolution of inflammation. ACKR2 is also involved in shaping chemokine gradients, ensuring proper immune cell trafficking and preventing chronic inflammatory conditions. Dysregulation of ACKR2 has been linked to autoimmune diseases, chronic inflammation, and cancer, where its altered expression may contribute to pathological immune cell infiltration and tumor progression. Its scavenging activity and role in regulating immune responses make ACKR2 a potential therapeutic target for inflammatory and immune-related diseases.
