Descripción
PHF1 is a chromatin-binding protein that contains a plant homeodomain (PHD) finger, allowing it to recognize methylated histone residues, particularly H3K36me3. PHF1 is part of the Polycomb repressive complex 2 (PRC2), which regulates gene expression through chromatin modification and silencing. It facilitates PRC2 recruitment to specific genomic loci, contributing to epigenetic regulation of development, differentiation, and proliferation. PHF1 is highly expressed in tissues with active epigenetic remodeling, such as embryonic stem cells and immune cells. Dysregulation of PHF1 has been associated with sarcomas and other cancers, where its aberrant activity affects transcriptional repression and genome stability. Knockdown studies reveal defects in PRC2-mediated H3K27 methylation, leading to disrupted chromatin states and altered gene expression profiles, highlighting its essential role in epigenetic regulation, cellular identity, and cancer development.
