| Assay Type | Activity: Active Biological Activity: A microchemotaxis assay was a carried out to determine the chemotactic effect of IL8 on Jurkat cells. 100 µm cell suspension (1 × 106 cells per ml) in RPMI 1640 FBS free medium were seeded to the upper chamber of a microchemotaxis system and Human IL8 (0.1 ng/ml, 1 ng/ml and 10 ng/ml dilutions in RPMI 1640) was added to the lower chamber with an 8 µm polycarbonate filter to separate the two compartments. The cells were incubated at 5% CO2, 37°C for 1 hour, the filter was removed and the cells in the lower chamber were observed at x100 magnification and counted at x400 magnification. CXCL8, also known as Interleukin-8 (IL-8), is a pro-inflammatory chemokine belonging to the CXC chemokine family. It plays a critical role in immune responses by promoting the recruitment and activation of neutrophils, which are essential in the body’s response to infection and injury. CXCL8 is secreted by a wide range of cells, including macrophages, epithelial cells, endothelial cells, fibroblasts, and keratinocytes, particularly in response to inflammatory stimuli like bacterial products, oxidative stress, and pro-inflammatory cytokines such as IL-1 and TNF-α. CXCL8 is a potent chemoattractant that directs the migration of neutrophils from the bloodstream to the site of infection or tissue damage. This chemotactic activity is critical for the rapid recruitment of neutrophils, allowing them to carry out their functions in pathogen destruction and the removal of damaged cells.
