Descripción
SH2D4A is an SH2 domain-containing protein that functions as a negative regulator of signaling pathways activated by receptor tyrosine kinases and cytokine receptors. It interacts with signaling molecules like STAT proteins and phosphatases to inhibit downstream pathways such as JAK/STAT and MAPK, modulating processes like cell proliferation, differentiation, and immune responses. SH2D4A is predominantly expressed in immune cells and tissues involved in hematopoiesis, where it plays a role in regulating cytokine signaling and immune homeostasis. It functions as a tumor suppressor in certain contexts, inhibiting excessive proliferation and survival signaling. Dysregulation or loss of SH2D4A has been linked to cancer progression and immune disorders due to uncontrolled activation of cytokine signaling. Knockout studies reveal hyperactive immune responses, abnormal proliferation, and defects in cytokine regulation, emphasizing its role as a critical negative regulator of cellular signaling pathways.
