Descripción

E2F4 is a member of the E2F family that predominantly functions as a transcriptional repressor to regulate genes involved in cell cycle arrest and differentiation. Unlike E2F1-3, which activate proliferation-related genes, E2F4 associates with pocket proteins such as p107, p130, and RB to repress E2F target genes during quiescence (G0 phase) and early G1. It is highly expressed in differentiated and non-proliferative cells, where it helps maintain cellular homeostasis and tissue stability. E2F4 is essential for hematopoiesis, where it promotes the differentiation of erythroid and myeloid cells by suppressing proliferation signals. Dysregulation of E2F4 has been linked to cancers and developmental disorders due to disrupted cell cycle arrest mechanisms. Knockout models exhibit defects in hematopoiesis, impaired differentiation, and embryonic lethality, underscoring E2F4’s critical role in maintaining cellular quiescence, differentiation, and growth suppression.
