DENN domain containing 4A (DENND4A) is a guanine nucleotide exchange factor (GEF) that plays a key role in activating Rab GTPases, specifically Rab10, which is central to vesicle trafficking, membrane remodeling, and exocytosis. DENND4A facilitates the GDP-to-GTP exchange on Rab10, enabling its active role in directing vesicles to the plasma membrane or specific intracellular compartments. Its regulatory functions are critical for receptor recycling, protein secretion, and cellular signaling pathways. DENND4A is expressed ubiquitously but exhibits higher expression in tissues with heightened vesicular transport demands, such as epithelial cells, neurons, and endocrine tissues. It also plays a significant role in maintaining endocytic recycling and cellular polarization, processes that ensure proper distribution of proteins and lipids within cells. Dysregulation of DENND4A has been linked to defects in membrane trafficking and signaling, contributing to conditions such as cancer, metabolic syndromes, and neurodegenerative disorders.
